I got Open up frame and i want to but wheelset. The questain is 29 or 27.5..
What is the actual naximum size of tries foe 29er wheelset? If i want to use 29er wheelset with not less 45mm tiers, what sort of wheelset can be Ok? How much slower can be 27.5 wheelset with 2.1 tires, compared with 29er with 40mm or 45mm?

40mm is the maximum for 700c (depending always a bit on tire and wheel model). the 27.5" tire is no slower than the 700c tire because they are the same diameter, that's the whole point. They are CALLED different diameters and they are different at the rim but they are the same in PHYSICAL dimension.
